Abstract Text: Research into GPI anchor protein deficiencies (APD), including paroxysmal nocturnal hemoglobinuria (PNH), often relies on custom or home-brew monoclonal antibody (mAb) cocktails, which can result in variability and inefficiency. Multicolor flow cytometry is critical for studying APD, including PNH, as it enables accurate identification and quantification of PNH clones across various cell lineages. In 2018, ICCS/ESCCA guidelines recommended standardized protocols for PNH assays, emphasizing the importance of reproducibility and accuracy.

To address these needs, we introduce the GPI-APD Kit with Veri-Cells™ Control, a fully standardized flow cytometry solution that ensures consistent and reliable assays for GPI-anchored protein deficiencies. The GPI-APD Kit includes the Veri-Cells positive control, which contains cells with undetectable GPI-anchored markers, ensuring reliable assay validation for PNH or other GPI-deficient cells. Additionally, the kit features FLAER conjugated to Spark Blue 488, a fluorochrome that highlights the absence of GPI-anchored surface markers, enhancing detection sensitivity. By incorporating a dried antibody assay format and a ready-to-use mAb cocktail, the kit improves staining reproducibility, minimizes preparation time, and reduces procedural errors.
